What is Sermorelin?
Sermorelin (GHRH 1-29 NH2) is a synthetic analogue of the first 29 amino acids of endogenous growth hormone-releasing hormone (GHRH). It was FDA-approved in 1997 for pediatric growth hormone deficiency and has an established safety record. In research and longevity contexts, it's used to stimulate natural GH release from the pituitary rather than administering exogenous GH directly — a significant distinction in terms of physiological mechanism and safety profile.
The key appeal of sermorelin over direct GH administration is that it works by stimulating the body's own pituitary to produce GH, maintaining the natural pulsatile release pattern and feedback regulation. This makes it a more physiologically conservative approach to GH optimization research.
How Sermorelin Works
Sermorelin binds GHRH receptors on pituitary somatotroph cells, stimulating synthesis and secretion of growth hormone. Crucially, this stimulation is subject to normal negative feedback — when GH and IGF-1 rise sufficiently, the pituitary becomes less responsive, preventing supraphysiological GH elevation. This self-regulating mechanism is absent with direct GH administration.
Research Findings
- GH restoration: Sermorelin effectively restores GH pulsatility in subjects with age-related GH decline (somatopause)
- Body composition: Consistent with GH effects — improved lean mass, reduced fat mass in research subjects
- Sleep quality: GH secretion peaks during slow-wave sleep; GH optimization research consistently shows improved sleep architecture
- IGF-1: Downstream IGF-1 elevation — the mediator of many GH effects on tissues
- Safety profile: Established safety record as an FDA-approved compound
Sermorelin vs CJC-1295
Both are GHRH analogues. Key difference: CJC-1295 (no DAC) has a similar short half-life to sermorelin but with greater binding affinity. CJC-1295 with DAC has a dramatically longer half-life (~7 days) enabling less frequent injection. For pulsatile GH research protocols, sermorelin or CJC-1295 (no DAC) paired with Ipamorelin is preferred over CJC-1295-DAC.
Dosing Protocols
Available in 2mg, 5mg, and 10mg vials. Standard protocol is subcutaneous injection before sleep to amplify the natural nocturnal GH pulse. Like all GHRH agonists, most effective when combined with a GHRP (like Ipamorelin) for synergistic GH release.
