What is NAD+?
NAD+ (nicotinamide adenine dinucleotide) is a coenzyme found in every living cell, where it plays a central role in energy metabolism and cell signaling. It exists in two forms: NAD+ (oxidized) and NADH (reduced), cycling between them as it transfers electrons in metabolic reactions — particularly in the mitochondrial electron transport chain where most cellular ATP is generated.
Beyond energy metabolism, NAD+ is a substrate for three major classes of enzymes involved in aging and longevity research: sirtuins (SIRT1-7), PARPs (poly ADP-ribose polymerases involved in DNA repair), and CD38/CD157 (involved in calcium signaling). The decline of NAD+ with age is now considered one of the hallmarks of cellular aging.
The numbers: NAD+ levels decline approximately 50% between ages 20 and 50, and continue declining. This decline correlates with mitochondrial dysfunction, reduced DNA repair capacity, and impaired sirtuin activity — all associated with aging.
Why NAD+ Declines with Age
Multiple mechanisms contribute to age-related NAD+ decline:
- Increased CD38 activity: CD38, an NADase enzyme, becomes more active with age and inflammation — consuming NAD+ faster than it can be synthesized
- Reduced biosynthesis: The salvage pathway (recycling NAM → NMN → NAD+) becomes less efficient with age
- Increased PARP activation: DNA damage (which accumulates with age) activates PARPs, which consume massive amounts of NAD+ in repair attempts
- Chronic inflammation: Inflammatory signaling increases NAD+ consumption through multiple pathways
Research Findings
- Mitochondrial function: NAD+ repletion research in aged mice restores mitochondrial function in skeletal muscle and other tissues to levels approaching younger animals
- Sirtuin activation: Sirtuins require NAD+ as a co-substrate. Restoring NAD+ levels reactivates sirtuin pathways associated with stress resistance, metabolic health, and DNA repair
- DNA repair: PARP-mediated DNA repair requires NAD+. Maintaining adequate NAD+ levels supports cellular ability to repair DNA damage
- Cognitive research: Several studies suggest NAD+ repletion may support neuronal function and mitophagy in neurodegenerative models
- Human trials: Growing body of human data showing oral NMN and NR raise blood NAD+ levels — direct NAD+ injection raises levels more acutely and predictably
Injectable NAD+ vs Oral Supplements (NMN/NR)
This is one of the most important distinctions in NAD+ research:
- Oral NMN/NR: Must be converted to NAD+ through the salvage pathway. Bioavailability is debated. Convenient, well-studied in human trials, reasonable evidence for raising blood NAD+ levels.
- Injectable NAD+: Bypasses gut and liver first-pass metabolism. Raises blood NAD+ more acutely and at higher peaks than oral equivalents. The bioavailability question is largely eliminated — what goes in gets absorbed. More expensive and requires reconstitution, but the research-relevant NAD+ elevation is more predictable and measurable.
For serious longevity research, injectable NAD+ provides more controllable, measurable variables than oral precursors. The tradeoff is cost and administration complexity.
Dosing Research Protocols
Injectable NAD+ dosing varies significantly by protocol:
- 500mg vials: Lower dose protocols, reconstituted in bacteriostatic water
- 1000mg vials: Standard research dosing
- 2g vials: Intensive protocols
- Administration: IV administration is used in clinical settings; subcutaneous is common in research protocols
- Frequency: Various protocols from daily to weekly depending on research goals
Where to Buy Injectable NAD+
Injectable NAD+ purity is critical — impurities in NAD+ preparations can cause significant adverse reactions. Domestic GMP manufacturing is the standard that matters here. UNYX Peptides carries 500mg, 1000mg, and 2g NAD+ vials, manufactured in their Arizona GMP facility.
- 100% USA-manufactured — GMP-certified Arizona facility (not imported from overseas)
