What is MOTS-c?

MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a mitochondria-derived peptide (MDP) — one of a class of small peptides encoded by mitochondrial DNA and secreted in response to metabolic stress. It was discovered relatively recently (2015) and has rapidly become one of the most exciting areas in longevity and metabolic research.

Unlike most peptides which are encoded in nuclear DNA, MOTS-c is encoded in mitochondrial DNA — specifically in the 12S ribosomal RNA gene. This evolutionary origin in the mitochondrial genome makes it fundamentally different from other research peptides, and its mechanisms reflect this: it functions as an intercellular and systemic signal of mitochondrial stress and metabolic state.

Why it's unique: MOTS-c is one of only a handful of bioactive peptides encoded in the human mitochondrial genome. Its discovery opened an entirely new field of mitochondria-to-nucleus and mitochondria-to-systemic signaling research.

The Discovery

MOTS-c was identified by Dr. Changhan David Lee and colleagues at USC Leonard Davis School of Gerontology in 2015. The initial paper in Cell Metabolism described how MOTS-c targets the folate cycle and de novo purine biosynthesis, activating AMPK — a master regulator of cellular energy homeostasis.

Subsequent research has expanded understanding of MOTS-c as a systemic peptide that circulates in blood and acts on skeletal muscle, adipose tissue, the brain, and other organs as a metabolic regulator and stress response signal.

Research Findings

Exercise Mimetic Properties

One of the most fascinating aspects of MOTS-c research is its exercise-mimicking properties. MOTS-c rises during exercise and appears to mediate some of the metabolic benefits of physical activity — improved glucose uptake, mitochondrial biogenesis, and fat oxidation. Research in sedentary aged mice showed that MOTS-c treatment partially recapitulated the metabolic benefits of exercise without the exercise.

This "exercise mimetic" potential makes MOTS-c particularly interesting for research on age-related metabolic decline, sarcopenia, and conditions where physical activity is limited.
